There are two major domains of interaction between the Escherichia coli release factors (RF-1 and RF-2) and each subunit of the ribosome. RF-2 has a binding domain on the shoulder and lower head region of the small subunit at the small lobe distant from the decoding site. This is in close proximity to one of the domains on the large subunit which includes the body dimer of L7/L12 and L11. The other domains of interaction, at the decoding site on the small subunit, and at the peptidyltransferase centre of the large subunit of the ribosome, are some distance from the first two, although the evidence for direct contact with the ribosome is less comprehensive. The release factors may therefore have two distinct structural domains, and in support of this concept RF-1 and RF-2 can both be cleaved into two fragments by papain. Region-specific antibodies, and antibodies against defined peptide within the RF sequences have given an indication that a significant part of an interacting RF molecule is in close proximity to the ribosome surface, confirming an observation by immunoelectron microscopy which suggested that the RF penetrates deeply into the cleft between the two subunits. A region of highly conserved primary sequence between the two release factors from E coli is also conserved in those from B subtilis suggesting it forms an important structural or functional domain. Antibodies against peptides from the N-terminal end of this region strongly inhibit binding of the RF to the ribosome.  相似文献

Eighty-five patients with Graves' disease in clinical remission after treatment for over 1 year by methimazole therapy (36 patients, group A) or subtotal thyroidectomy (49 patients, group B) who became undetectable for serum thyrotropin levels (TSH less than 0.05 mU/l), were further followed for 1 year or more. Eight patients in group A (22%) and 7 patients in group B (14%) relapsed. Eleven patients in group A (30%) and 5 patients in group B (10%) had fluctuating patterns of free T4 in the upper normal to slightly supranormal range indicative of subclinical hyperthyroidism. The remaining patients continued to have undetectable TSH levels or restored normal TSH levels and normal thyroid hormone concentrations in sera. The results of the present study indicate that the occurrence of undetectable serum TSH concentrations in Graves' disease patients previously treated with methimazole or surgery are not necessarily predictive of clinical relapse because the eventual outcome is variable.  相似文献

Advances in the understanding of Ras oncoprotein function suggest novel points for anti-tumor intervention. First, upstream-acting guanine nucleotide exchange factors and SH2/SH3 domain-containing adaptor proteins that link Ras with growth factor receptor tyrosine kinases have recently been characterized. Second, work on downstream-acting Ras effector functions including the Ras GTPase-activating protein (p120GAP) and the Raf kinase has revealed direct biochemical interactions that are functionally required for oncogenic Ras signalling. We summarize progress in these areas and discuss the potential for novel applications to anti-cancer chemotherapy.  相似文献

An essential part in the development of informative linkage maps is to include genetic markers that have been anchored by physical mapping. Here a set of 18 porcine cosmid-derived genetic markers are reported that have been mapped by linkge analysis, and that also have been physically localized by fluorescence in situ hybridization (FISH). Three different strategies were used to establish polymorphic markers from the cosmid clones. Firstly, dinucleotide microsatellite loci were derived by sequencing cosmid subclones containing (CA), repeats. Secondly, variable SINE 3′ poly(A) tracts (SINEVA) were identified by direct SINE-PCR amplification of cosmid clones. Thirdly, the cosmids were used in Southern blot hybridization to detect restriction fragment length polymorphisms (RFLPs). Compared with the most recent consensus compilation of the porcine gene map, the present assignment of markers to chromosomes Zp, 3, 4, 10, 12q, and 16 represents the first loci mapped to these chromosomes, for which linkage as well as in situ data are now available.  相似文献

Chrysophtharta bimaculata (Olivier) (Coleoptera: Chrysomelidae) is a major defoliator of regeneration eucalypt trees in Tasmania causing a significant reduction in height and diameter increment of trees which reduces wood volume per hectare. A study to conserve and enhance the efficiency of coccinellid species chieflyCleobora mellyi (Mulsant) (Coleoptera: Coccinellidae), and the cantharid,Chauliognathus pulchellus (Macleay) (Coleoptera: Cantharidae), for the biological control ofC. bimaculata was conducted in young regeneration forests in southern Tasmania from 1991–92. Cantharid adults and coccinellid adults and larvae feed onC. bimaculata eggs and, to a lesser extent, young larvae. The study found that coccinellids were more active throughout the egg and early (1st and 2nd) stage ofC. bimaculata. The cantharid, however was active only during the egg stage of the prey and then disappeared from the plantation. The coccinellids were therefore the most useful predators, but their population declined when the prey reached the 3rd and 4th stages. As shortage of food may account for this decline, supplementary food was provided in the form of sucrose sprays or sugar granules at a feeding station. This resulted in the retention of both predators and particularly the coccinellids and enhanced their efficacy.  相似文献

We have examined the ability of insulin and dsRNA, a well-known interferon inducer, in relation to their effects on cell growth, to induce the expression of hsp 70 and the synthesis of interferon in epithelial HT-29 and fibroblast Madin-Darby bovine kidney (MDBK) cells. Insulin was mitogenic in both MDBK and HT-29 cells; MDBK cells nevertheless required much higher concentrations. DsRNA stimulated the growth of MDBK but inhibited that of HT-29 cells. Both substances induced a transient synthesis of hsp 70 in HT-29 and MDBK cells with similar kinetics. However, whereas both insulin and dsRNA efficiently induced 2′5′ oligoadenylate synthetase and an antiviral state through interferon synthesis in HT-29 cells, only dsRNA caused these effects in MDBK cells. Thus, insulin cannot, unlike dsRNA, elicit an antiviral state in all cell systems, although, like dsRNA, it can induce hsp 70, thereby suggesting the cell specificity of insulin action. These results reveal that the mitogenic and IFN-inducing effects of insulin and dsRNA are dependent on the cell type and unrelated to hsp 70 expression.  相似文献

Fertile eggs obtained from alligators reared in captivity typically exhibit high rates of embryonic mortality. Also, the fatty acid composition of the yolk lipid of the captive eggs is markedly different from that observed in eggs from wild alligators, possibly as a result of differences in maternal diet in the two situations. The fatty acid compositions of tissue lipids during the embryonic development of wild and captive alligators were compared. The lipids of liver, adipose tissue and heart of the two types of embryo displayed fatty acid profiles which generally reflected the acyl compositions of the respective yolks. Thus the lipids from these tissues of the captive embryos contained markedly higher proportionate levels of linoleic and linolenic acids, lower levels of palmitoleic acid, and, in general, lower levels of docosahexaenoic acid and other C20 and C22 polyunsaturates, in comparison to the values for the wild embryos. In contrast, the fatty acid composition of the brain phosphoglycerides was very similar in the two types of embryo. Thus, at least in those embryos which had survived during the developmental period studied, the brain was able to maintain a relatively constant fatty acid composition, in spite of major differences between the wild and captive eggs in the proportions of the various fatty acids supplied from the yolk. It is suggested that a major cause of embryonic mortality in the captive embryos could be a failure to maintain an adequate level of docosahexaenoic acid in the developing brain.  相似文献
